No, most of the Shimano Road rear derailleurs have a maximum tooth count of 27 or 28 teeth, so the 36 tooth would be pretty far out of the question. If people want to run a cassette of this size, they typically choose a mountain derailleur instead of a road one.

My rig is on a Co-Motion tandem. I have Ultegra STI 10 speed shifters, a Shimano XT M771 derailleur, and this SRAM cassette.
Simple to tune. Works just great.
