The R540 is a 3-bolt SPD-SL cleat, this adapter is for using 2-bolt SPD cleats to shoes already set up for 3 bolt cleats. If your shoes have a 3-bolt sole you don't need any adaptors.

Most likely not, the plates are designed for shoe made around 2000-05

They wont because of the sole shape. They only work with the older Genius and Millennium shoes
